Williams, a seven-time Miami champion, moved on with a 6-1, 6-1 win over fellow American CiCi Bellis.
Svetlana Kuznetsova, the No. 24 seed, is next up for Williams after dispatching 13th-seeded Angelique Kerber 6-3, 3-6, 6-3.
Third-seeded Simona Halep continued her strong start to the 2015 season with a 6-4, 7-5 third-round victory over No. 30 seed Camila Giorgi.
No. 15 seed Flavia Pennetta will face Halep in the fourth round thanks to her 7-6 (7-5), 7-6 (8-6) win over Victoria Azarenka.
In a minor upset, 27th-seeded Sabine Lisicki fought past fifth-seeded Ana Ivanovic 7-6 (7-4), 7-5. Lisicki will next meet No. 11 Sara Errani, who defeated Garbine Muguruza 4-6, 6-4, 6-1.
American Sloane Stephens beat Johanna Larsson 6-4, 6-4 and will face Belinda Bencic, who got past Tatjana Maria 6-4, 7-5.
The 2015 Miami champ will earn $900,400.
