Poland's Radwanska, the reigning champion at this event, was dominated in a 7-5, 6-2 loss to American Bethanie Mattek-Sands on the hardcourts at NSW Tennis Centre.
Second-seeded Czech Petra Kvitova blitzed American qualifier Christina McHale 6-1, 6-0, and fifth-seeded German Angelique Kerber had little trouble in a 6-3, 6-4 victory over Estonian Kaia Kanepi.
American Madison Keys was a 6-0, 3-6, 7-6 (7-3) winner against Ajla Tomljanovic of Croatia, and Spain's Carla Suarez Navarro grabbed a 7-6 (7-4), 6-3 win against Ekaterina Makarova of Russia.
