Jake Peavy (11-11) gave up just two runs on four hits with a walk and six strikeouts over six innings to get the win.
"That's a big one we have tomorrow and I hope the fans are out there for us," said Peavy. "A win tomorrow would be huge for us then we head to Kansas City so we have our work cut out for us."
Scott Diamond (11-8) gave up six runs on seven hits in 5 1/3 innings to lose his second straight start. Joe Mauer hit a solo home run in the loss for the Twins, who have lost five of six.
"I feel like I've been battling a lot lately and today was no different," said Diamond. "I felt a little better today and I did make some pitches, but they put some good swings on them."
Mauer's 10th homer of the season came in the first inning to give the Twins the lead, but Chicago answered in the third.
With men on first and third, Wise's sac fly scored one and Youkilis followed with a double to plate another. Viciedo's RBI double in the fourth gave the White Sox a 3-1 lead.
The Twins got one back in the fifth on consecutive doubles from Trevor Plouffe and Alexi Casilla, but Chicago blew the game open in the sixth.
Dunn led off the frame with his 39th of the season and, after Alex Rios tripled, Viciedo launched a pitch over the wall in left for his 21st of the year.
That was it for Diamond as Anthony Swarzak took the mound, but he gave up a sacrifice fly to Wise and RBI singles by Youkilis and Dunn as the White Sox took a 9-2 advantage.
