Barthel blew past Dutchwoman Kiki Bertens 6-3, 6-2 at Bastad Tennis Stadium.
A Day-1 upset came when Ukrainian Lesia Tsurenko dismissed eighth-seeded Spaniard Carla Suarez Navarro 7-5, 6-0, following a rain delay.
This week's top seeds are Sara Errani and German Julia Goerges. The surprise French Open runner-up Errani is fresh off her clay-court title in Palermo in her native Italy last week.
Slovenian Polona Hercog beat Sweden's own Johanna Larsson in last year's Bastad finale.
